Background

ALP is a hydrolytic enzyme with specificity for phosphate substrates, which can catalyze the hydrolysis or dephosphorylation of a variety of phosphorus-containing compounds in alkaline media. In biological analysis, ALP can be used as an enzyme label or indicator in a reaction to quantify the combined process of immunoassay and genetic testing or other methods. At present, the detection methods of ALP activity mainly include colorimetric method, fluorescence analysis method, electrochemical analysis method and surface enhanced Raman scattering method. Highly sensitive detection of ALP

Hydrogen peroxide (H2O2) is catalyzed by horseradish peroxidase (HRP) to produce hydroxyl groups, which can oxidize o-DHB to o-benzoquinone and quench GQDs fluorescence. Ascorbic acid 2-phosphoric acid (AAP) can be catalyzed by ALP to produce ascorbic acid (AA). AA with strong reducing ability can effectively remove H2O2 and hydroxyl in solution, and then inhibit the formation of o-quinone, inducing the fluorescence recovery of GQDs. As the activity of ALP increased, the fluorescence at 440 nm increased continuously, so a highly sensitive detection method for ALP was established.
